JOB SUMMARY:

Research technician to assist with molecular and developmental biology studies of butterfly adaptation. Primary duties of this position will include assisting with generating DNA/RNA sequencing libraries, CRISPR mutagenesis experiments, management of butterfly rearing facilities, and basic lab management. Other duties as assigned.

JOB DUTIES:

30% - Essential - Work independently to generate DNA/RNA sequencing libraries: DNA/RNA extraction using Trizol and/or DNA lysis buffer and columns. Physical DNA fragmentation and/or base hydrolysis fragmentation of RNA. Custom and kit-based sequencing library production, including DNA-DNA and possibly DNA-RNA ligations, sample indexing, etc. SPRI bead DNA sequencing library size selection, cleanup, and submission for sequencing. Requires molecular biology experience.

30% - Essential - Work independently to perform CRISPR/Cas9 mutagenesis experiments: Design sgRNAs targeting coding and non-coding DNA sites. Pool sgRNAs and perform microinjections in collected eggs. Screen potential CRISPR mutants for phenotypes. DNA extraction and PCR validation of CRISPR mutagenesis experiments. Identification and Interpretation of mutant electropherograms. High molecular weight DNA extraction and long-read sequencing for further mutagenesis validation. Requires developmental and/or molecular biology experience.

20% - Essential - Management of butterfly rearing facilities: Monitor and maintain schedules for host and nectar plant watering, caterpillar feeding, cage maintenance, etc.

20% - Essential - Basic lab management: Maintain list of chemicals/reagents, keep track of orders and shipping, safety documentation, etc.

MINIMUM REQUIREMENTS:

Education - High School or Equivalent and journey-level technical experience in a related laboratory setting.

Work Experience 0+ years

PREFERRED REQUIREMENTS:

Education - Bachelor's Degree - Biology, evolutionary biology, developmental biology, genetics, or a related field.

Work Experience 1+ years with molecular biology, developmental biology, or genomics is preferred.

No Budget Responsibilities - No fiscal responsibility for the department's budget.

PHYSICAL REQUIREMENTS:

50% - Sit (stationary position) for prolonged period

20% - Walk or move about

70% - Use hands or feet to operate or handle machinery, equipment, etc

25% - Communicate, converse, give direction, express oneself

50% - Recognize or inspect visually

10% - Move, transport, raise or lower

50% - Perceive, observe, clarity of vision

WORKING CONDITIONS:

05% - Wet or humid

70% - Chemical hazards

10% - Exposure to standard molecular biology lab hazards, will NOT work with vertebrate or prokaryotic samples. Butterfly rearing facilities are within a greenhouse. Other exposure: butterflies.

WORK SCHEDULE:

Standard Hours: 37.5

COMPENSATION INFORMATION

Anticipated Salary Range: $36,800 - $41,500

Salary is dependent upon several factors including, but not limited to, a candidate's previous experience, knowledge, skills and performance in accordance with Clemson's compensation guidelines.

ESSENTIAL PERSONNEL LEVEL

Normal Operations

Required to follow emergency facility closure directives, and not normally expected work on-site during emergency situations.
